Flavin radicals, conformational sampling and robust design principles in interprotein electron transfer: the trimethylamine dehydrogenase-electron-transferring flavoprotein complex.

چکیده
TMADH (trimethylamine dehydrogenase) is a complex iron-sulphur flavoprotein that forms a soluble electron-transfer complex with ETF (electron-transferring flavoprotein). The mechanism of electron transfer between TMADH and ETF has been studied using stopped-flow kinetic and mutagenesis methods, and more recently by X-ray crystallography. Potentiometric methods have also been used to identify key residues involved in the stabilization of the flavin radical semiquinone species in ETF. These studies have demonstrated a key role for 'conformational sampling' in the electron-transfer complex, facilitated by two-site contact of ETF with TMADH. Exploration of three-dimensional space in the complex allows the FAD of ETF to find conformations compatible with enhanced electronic coupling with the 4Fe-4S centre of TMADH. This mechanism of electron transfer provides for a more robust and accessible design principle for interprotein electron transfer compared with simpler models that invoke the collision of redox partners followed by electron transfer. The structure of the TMADH-ETF complex confirms the role of key residues in electron transfer and molecular assembly, originally suggested from detailed kinetic studies in wild-type and mutant complexes, and from molecular modelling.

Small angle x-ray solution scattering has been used to generate a low resolution, model-independent molecular envelope structure for electron-transferring flavoprotein (ETF) from Methylophilus methylotrophus (sp. The isolation and partial characterization of a flavoprotein which functions as the electron acceptor of trimethylamine dehydrogenase (EC 1.5.99.7) from a methylotrophic bacterium is described. It has a molecular weight of 77,000 and is composed of two dissimilar subunits. All preparations examined contained only 1 mol of FAD/m01 of the flavoprotein. Trimethylamine dehydrogenase, in the presenc...

The interaction between two physiological redox partners, trimethylamine dehydrogenase and electron-transferring flavoprotein, has been characterized quantitatively by analytical ultracentrifugation at 4 degrees C. Analysis of sedimentation-equilibrium distributions obtained at 15 000 rpm for mixtures in 10 mM potassium phosphate, pH 7.5, by means of the psi function [Wills, P. R., Jacobsen, M....
متن کاملThe electron-transferring flavoprotein as a common intermediate in the mitochondrial oxidation of butyryl coenzyme A and sarcosine.
Butyryl coenzyme A dehydrogenase and a transfer protein required for maximal activity as measured by reduction of 2,6-dichloroindophenol have been solubilized from monkey (Macaca mulatta) liver mitochondria by sonic irradiation. The dehydrogenase has been purified approximately 60-fold and identified as a flavin adenine dinucleotide-specific flavoprotein. The required protein has been found to ...
متن کاملThe Electron-transferring Flavoprotein of Primate Liver Mitochondria. Purification of the Enzyme and Reconstitution of the Sarcosine Oxidase System.
The oxidation of sarcosine (N-methylglycine) by purified rat liver sarcosine dehydrogenase is known to require the participation of a flavin adenine dinucleotide-dependent protein, the electron-transferring flavoprotein, for efficient transfer of electrons between reduced dehydrogenase and 2,6-dichlorophenolindophenol (l), or cytochrome c (a), or cytochrome b (2). On the basis of spectral evide...
